Not with the standard front and rear sprockets, no. Anyhow you can't install a 6'th gear anyway, but if you could, then you could change the ratio with other sprockets.
By doing so, you could make the 6'th gear so that the engine would rev the same as it would in standard trim in 5'th, this giving you a first gear that could pull the @ up along the northern side of K2.
If you're into different gear ratios, you could investigate other Honda bikes using basicly the same engine, like the Hawk GT, or BROS 650, or Transalp 600-700, perhaps there are others too like the NTV650.
But I doubt if the result will justify the effort in the end.

Yes it is possible.
You need to design and manufacture a new selector drum, gearbox shafts, gears, selector arms etc. It is probably that it can be done without any crankcase mods.
I'd hate to think of the cost of designing and manufacturing a gearbox from scratch. Just one gear can cost £100 from Honda - and that's a mass production price!

Rather than re-engineering a gearbox, try putting a larger front sprocket on if you want to overdrive the engine. They cost around £6 so you could afford to play around with ratios. With a bit of simple maths you can work out the rpm difference it'll make for a given sprocket. You wouldn't get the spread of ratios that you'd get from an extra gear but it'd at least tell you whether it's worth taking further.

Fitting an additional gear ratio and re-engineering the box to turn it into a 6 speed box is very different from simply putting taller gearing on a bike with a 5 speed box. With a six speed box, you can have a lower first and a taller top gear rather than one or the other.
It would be a great mod if it was practical and something that I would have liked on my RD04. It could have done with a lower first for some off-road stuff and a taller 5th for motorways.
Of course, if you use some cheap and nasty no-name £6 sprocket, you'll be needing to replace the output shaft at some point anyway so that may be a good start point.
